DNA content measurement for DNA ploidy and cell cycle analysis.
نویسندگان
چکیده
This unit presents the most common methods of DNA content analysis. The techniques differ primarily in the mode of cell permeabilization (detergents versus prefixation with alcohol) and composition of the stain solution. Protocols are given for analysis of fixed cells, tissue samples, live cells (for subsequent sorting and culture), archival paraffin-embedded tissue, and apoptotic cells.

A survey on the DNA content in Transitional cell carcinoma of bladder and its relation with histological graging
Transitional cell carcinoma(T.C.C) of bladder display an upredictable biological behavior.Morphologic methods of grading this tumor are insuficient to predict the clinical outcome of the patients.the aim of this study was to investigate the relationships between histological grading staging and DAN ploidy especially in intermediate grade II(IIa and IIb). in a retrospective study of tissue spec...
متن کاملNUCL EAR DNA CONTENT AND DNA PLOIDY ANALYSIS IN BREAST CANCER
To investigate the patterns of DNA ploidy and proliferating activity in breast cancer and relate them to other prognostic factors, paraffin blocks of 53 cases of breast carcinoma were studied. Cancer cells obtained by mechanical tissue disaggregation were examined for DNA content, ploidy and S-phase fraction. DNA assay was done using a CAS interactive image analyzing system. All of the cas...
متن کاملاثرات گاز کلر بر سیکل سلولی و محتوی DNA گلبولهای سفید موش سوری The Effects of Chlorine on Cell Cycle and DNA Content of WBC in Mice
Background & Aim: Chlorine has been known as a mucus membranes and respiratory tract irritant. This gas can increase free radicals which cause cell damage. The aim of the present study was to measure DNA content and cell cycle in white blood cells after chronic chlorine poisoning. Materials & Methods: A clinical experimental study was carried out on 80 male mice(40 mice as sample and 40 as ...
متن کاملSimultaneous analysis of cell cycle kinetics at two different DNA ploidy levels based on DNA content and cyclin B measurements.
The methods of cell cycle analysis that rely on DNA content measurements cannot discriminate between cells at different phases of the cycle if these cells have similar DNA content. This limitation can be circumvented by measurement of another cell cycle phase-specific cell constituent in addition to DNA content, followed by bivariate analysis of the correlated data. The aim of the present study...
متن کاملDNA Ploidy and Cell Cycle Analysis in Cancer Diagnosis and Prognosis
During the past 10 years, there has been considerable interest in the application of new technologies to identify human malignancy and predict disease outcome. Markers of cell proliferation and the techniques of flow cytometry and image analysis for the determination of DNA total content in human tumor cells have been at the forefront of these new developments. This review will focus on the cli...
